HIE, being a rare disease, has long been the subject of research interest, but the difficulty of treatment and limited therapeutic options have led to a lack of effective intervention methods for an extended period. Hypoxic-ischemic encephalopathy (HIE) is one of the primary causes of acute neonatal death and chronic neurological damage, and severe cases of HIE can result in secondary sequelae such as cognitive impairment and cerebral palsy. However, currently, effective intervention measures are still lacking. In this study, the research and development team at Beijing Bao Feng successfully mitigated brain damage in HIE rats and significantly improved their cognitive abilities by administering Acer Truncatum Seed Oil (ASO) for 30 consecutive days. The results observed through lipidomics analysis showed a decrease in unsaturated fatty acids and an increase in lyso-phospholipids in the brains of HIE rats. However, after 30 days of ASO treatment, there was an increase in phospholipids, phosphatidylcholines, and unsaturated fatty acids in both the serum and brain tissue, while lyso-phospholipids and oxidized glycerophospholipids decreased. Enrichment analysis indicated that ASO intake mainly affected sphingolipid metabolism, fat digestion and absorption, glycerolipid metabolism, and glycerophospholipid metabolism pathways in the serum and brain. Cluster analysis, correlation analysis, and confirmatory factor analysis further confirmed that the cognitive improvement after ASO treatment could be attributed to the increase in essential phospholipids and ω3/6/9 fatty acids in the brains of HIE rats and the decrease in oxidized glycerophospholipids. These research findings suggest that ASO holds the potential to become an effective food supplement for infants with ischemic-hypoxic conditions.
